We generated a total of 237.7× sequence coverage of raw ultralong ONT data and 69.4× coverage of PacBio HiFi data for assembling the maize Mo17 genome (Fig. T2T assembly of all ten chromosomes of the maize Mo17 genome Here we report the complete telomere-to-telomere (T2T) Mo17 genome using a combination of ultralong Oxford Nanopore Technology (ONT) and PacBio HiFi reads, which marks a major step forward for genome assembly and uncovers the recalcitrant structural features of the highly complex maize genome. Yet, with the exception of B73-Ab10 with 53 gaps, all other assembled maize genomes reported still have hundreds or thousands of unfilled gaps. More recently, high-quality genome sequences of 25 nested association mapping (NAM) founder lines and a further improved B73 assembly (version 5) have also been reported 37. 35) and the inbreds underlying classical genetic studies W22 (ref. 34), the genetic transformation competent line A188 (ref. 30), the tropical lines SK 31 and K0326Y 32, European inbred lines F7, EP1, DK105 (ref. Due to the rich history of genetic studies in maize and its exceptional intraspecific genome diversity 26, 27, 28, 29, several additional inbred lines have also been sequenced, including the key Iodent line PH207 (ref. Therefore, the generation of their genome sequences is of great significance 8, 12, 13. The inbred lines B73 and Mo17 are the parental lines of one of the best-performing early commercial hybrids and of the most widely used bi-parental genetic mapping population 24, 25. Having a genome size very close to that of humans, while containing over 80% repetitive sequences, maize is also known as a model for complex genomes. Similarly, an essentially completed banana genome of 485 Mb has been reported 22, while the watermelon genome, with a size of 369 Mb and only several dozens of 45S rDNA copies, has been completely assembled 23. For the relatively compact Arabidopsis 18 and rice 19, 20, 21 genomes, high-quality gap-free reference genomes have also been achieved, with only several chromosomal ends, including nucleolus organizer regions (NORs), remaining incomplete. The assembly of the entire chr3 and chr9 of the maize line B73-Ab10 has also been reported 17. Benefitting from sequencing technology advances, the entire human X chromosome and chromosome 8 (chr8) have been completely assembled from telomere to telomere without gaps 14, 15, and a completely assembled human genome has recently been released 16. Using PacBio sequencing, both the improved B73 genome 12 and the Mo17 genome 13 achieved a contig N50 of more than 1 Mb, leaving only a few thousand gaps for this highly complex crop genome. The development of long-read, single-molecule DNA sequencing technologies substantially improved genome assembly quality 9, 10, 11. For instance, the first reported maize B73 inbred line genome had more than one hundred thousand gaps, with each tiling bacterial artificial chromosome sequence on average having more than ten gaps 8. Due to cost and read-length limitations, these earlier reported draft genomes typically had tens of thousands of gaps in the pseudochromosomes. All these pioneering genome sequencing projects were based on classical Sanger sequencing technology. 1), numerous draft genomes with varied extents of completeness have been reported, including fruit fly 2, Arabidopsis 3, human 4, 5, mouse 6 and the model crop species rice 7 and maize 8. Followed by the decoding of the very first eukaryotic genome, the 12 Mb yeast nuclear genome in 1996 (ref. Genome sequencing has been fundamental for the advancement of many aspects of basic biology as well as medical and agricultural applications. The complete Mo17 genome represents a major step forward in understanding the complexity of the highly recalcitrant repetitive regions of higher plant genomes. Additionally, complete assemblies of all ten centromeres enabled us to precisely dissect the repeat compositions of both CentC-rich and CentC-poor centromeres. The assembly of the entire nucleolar organizer region of the 26.8 Mb array with 2,974 45S rDNA copies revealed the enormously complex patterns of rDNA duplications and transposon insertions. There were several super-long simple-sequence-repeat arrays having consecutive thymine–adenine–guanine (TAG) tri-nucleotide repeats up to 235 kb. The 2,178.6 Mb T2T Mo17 genome with a base accuracy of over 99.99% unveiled the structural features of all repetitive regions of the genome. Through generating deep coverage ultralong Oxford Nanopore Technology (ONT) and PacBio HiFi reads, we report here a complete genome assembly of maize with each chromosome entirely traversed in a single contig. A complete telomere-to-telomere (T2T) finished genome has been the long pursuit of genomic research.

We include specific suggestions for how VSEPR-style rationalizations of molecular structure can be replaced with more accurate conceptions of hybridization and its relationship to electronegativity and molecular geometry, in accordance both with Bent's rule and the consistent features of modern wavefunctions as exhibited by natural bond orbital (NBO) analysis. VSEPR-style conceptions thereby detract from more accurate introductory-level teaching of orbital hybridization and bonding principles, while also requiring wasteful ''unlearning'' as the student progresses to higher levels. Using VSEPR theory, we predict that the two regions of electron density arrange themselves on opposite sides of the central atom with a bond angle of 180. VSEPRTheory(MolecularShapes) the central atom, X an atom bonded toA, E alonepair on A pairs on X or other atoms, but we dont care. Step 2: Determine the Central Atom of the Molecule. VSEPR-style representations of orbital shape and size are shown to be fundamentally inconsistent with numerous lines of experimental and theoretical evidence, including quantum mechanical ''symmetry'' principles that are sometimes invoked in their defense. total valence electron number in CO32- is 4 + 63 + 2 24. s orbitals are spherically symmetric around the nucleus - in each case, like a hollow ball made of rather chunky material with the nucleus at its centre. We describe the logical flaws, experimental contradictions, and unfortunate educational repercussions of common student misconceptions regarding the shapes and properties of lone pairs, inspired by overemphasis on ''valence shell electron pair repulsion'' (VSEPR) rationalizations in current freshmanlevel chemistry textbooks. The "s" tells you about the shape of the orbital. The "1" represents the fact that the orbital is in the energy level closest to the nucleus. The orbital occupied by the hydrogen electron is called a 1s orbital. Our chemistry teacher told us that both VSEPR theory (which says that the electron pairs in the valence shell of an atom arrange themselves in such a way that repulsions among them are minimized and this arrangement of the electron pairs determines the shape of a particular molecule) and hybridization (which is the intermixing of a particular nu. You can think of an orbital as being the region of space in which the electron lives. VSEPR theory predicts the shapes of molecules, and hybrid orbital theory provides an explanation for how those shapes are formed. (b) Two of the hybrid orbitals on oxygen contain lone pairs, and the other two overlap with the 1 s orbitals of hydrogen atoms to form the OH bonds in H 2 O. BondingMOs have lower energy than the AO forming. It specifically uses the Coulombic repulsion between electrons as a basis for predicting electron arrangement. Such a region of space is called an orbital. (a) A water molecule has four regions of electron density, so VSEPR theory predicts a tetrahedral arrangement of hybrid orbitals. VSEPR theory (Valence Shell Electron Pair Repulsion theory) states that electrons repel each other because they are all negatively charged, so the bonds and lone pairs in a molecule will get as. Molecular Orbital (MO) Theory Total number of MOs formed, must be equal to that of AO combined to make them. The Valence-Shell-Electron-Pair-Repulsion (VSEPR) theory can predict molecular geometry by minimizing electron-electron repulsion. First, write the Lewis structure of the molecule, in this model, consider only the electron pairs of the central atom. The diagram shows a cross-section through this spherical space.ĩ5% of the time (or any other percentage you choose), the electron will be found within a fairly easily defined region of space quite close to the nucleus. Here are some guidelines for applying the VSEPR model: 1. In the hydrogen case, the electron can be found anywhere within a spherical space surrounding the nucleus. You keep on doing this over and over again, and gradually build up a sort of 3D map of the places that the electron is likely to be found. You have no idea how it got from the first place to the second. Soon afterwards, you do the same thing, and find that it is in a new position. Suppose you had a single hydrogen atom and at a particular instant plotted the position of the one electron. Read More: Is The Current Gen Silverado Worth Buying? Aerodynamics were the name of the game back then, and GM certainly understood the assignment. Additionally, this truck would be the very first General Motors truck to be flow tested in a wind tunnel, which is why you see a lot of those really sweeping body lines and curved panels in comparison to trucks before that. No longer were your windows just sheets of flat glass that you just rolled up and down in the doors. Beyond the beloved - or maybe maligned - square wheel wells that these pickups featured, they came with a completely restyled design, with more rounded features and seats that didn't completely suck when you wanted to drive it for more than 35 minutes.įun fact, also - did you know that even though we call them square-body Silverados, this generation was actually introduced as the round body generation of CNK pickups? This is because for the first time in the history of pickup trucks, GM would introduce a truck with curved-side window glass. The brand new square body pickup truck would go onto set the benchmark for the next 30 years of GM's pickup truck production, at least in terms of exterior styling. Because of this, GM would go to the drawing board and come back with a design so iconic, so classic, and so good-looking that they would leave it relatively unchanged for the next 15 years. After the introduction of their second generation of the C/K line of pickup trucks, GM knew that their new truck had to be an absolute banger if they wanted a shot at the top of the food chain when it came to truck sales in America. In 1973, things were heating up for the old General Motors Corporation. Top Design Features of Square Body Chevrolet Silverado Trucks If you haven't guessed by now, we're answering the question "Is a Square-Body Chevy Worth It?" so let's jump right in and show you! Today, we'll be talking about those oh so fantastic square wheel wells, the boxy grille headlight design, and the introduction of the very first dual-wheel pickup in existence. In the same way, there are plenty of trucks out there that get a lot of hype, but there are few that live up to the legacy of the square body Chevy. Look at Babe Ruth - dude played in the 1920s and is still getting talked about to this day, and he's even got a candy bar named after him, which is something we bet you can't match. Heroes get remembered, but legends never die. Recall that we already have another drawing lesson about pick-up – how to draw a Chevy Truck. Here we need to draw wheels that look like ovals (or flat cylinders, depending on the point of view). This step is approximately the same in all drawing lessons about cars (from Honda to Ferrari). Using straight lines draw out the pickup box with a rear bumper, rear lamps and wheel arch. Here we do about the same thing as we did in the previous step. Carefully draw out the door, a handle, moldings and the front wheel arch. Now let’s move to the lateral part of our Chevrolet Pickup. Draw out the rear-view windows and move to the next step. Draw out the windscreen and side windows. Let’s move to the cabin of our Chevrolet Silverado. Also using straight and clear lines draw out the details of the bumper. Let’s move to the hood by drawing it with straight and clear lines. In the center of the radiator grill we draw the Chevrolet logo. On both sides of the radiator grille we draw out the headlights.
